百人町三丁目遺跡は武蔵野台地の東部、神田川と妙正寺川がかつて合流していた場所の南西側の豊島台に位置する。　百人町三丁目付近は江戸時代には大久保百人組屋敷があった場所の一部にあたり、現在の町名もこの百人組に由来している。発掘調査が行われた場所は、大久保百人組屋敷の北通りに面した場所で、屋敷があった場所と考えられる。調査の結果、江戸時代と近代以降の遺構と遺物が確認された。江戸時代の遺構のうち、長軸方向が南北で、南側の道路に直交する方向で南北一直線に並ぶ長方形の土坑とそれに並行する溝状遺構を確認した。この土坑と溝状遺構については屋敷境もしくはそれに関連する遺構の可能性が考えられる。またこの土坑は、つつじの栽培を列植した、垣根や屋敷の構などの可能性も考えられる。その他の土坑のうち、平面形が円形で、比較的大型の土坑と、平面形が不整形で底面に凹凸が認められるものの多くは植栽痕と考えらる。
