• Note (General): type:text; 貝類は多様な色彩の貝殻を持つが、その色素はいまだ化学的に同定されておらず、生合成経路や役割、進化的な背景など、多くは解明されていない。本研究では緑色の貝殻をもつミドリイガイに含まれる青色色素に着目した。これまでに青色色素はフェノキサジンを発色団にもつ色素ペプチドであることが分かっているが、その立体化学が推定に留まっている。本化合物の完全な構造決定を目指し、モノトリと合成の両面から取り組んだ。 江ノ島にてミドリイガイを採取し、その色素を抽出した。抽出物中の青色色素をHPLCで定量すると、その含量は過去に得た抽出物よりも少なく、約20%しか含まれていなかった。ミドリイガイの貝殻の色には個体差があり、鮮やかな緑色とくすんだ緑色の個体が存在する。個体群により青色色素量に差があることが疑われた。そこで、江ノ島、金沢漁港、深浦漁港の3地点で採取した個体群を用いて、画像解析と青色色素の定量を行い、両パラメータ間の関係を調べた。RGB分解の結果、江ノ島群は有意に赤色要素の割合（R%）が多く、G%とB%の要素が少ないことが分かった。他群では逆にR%が少なく、G%とB%の要素が多かった。青色色素定量の結果、江ノ島群の青色色素収量は他群の10–40%ほどと少なかった。青色色素量はR%と負の相関を、B%と正の相関を示した。外観上、赤みが少なく、より青い個体を使用することで青色色素を多く得られることが分かった。抽出物を精製し、約80 μgの青色色素を単離した。X線結晶構造解析による構造決定を目指し、蒸気拡散法による結晶化を検討した。薄膜状および粒状の析出物が得られたものの、結晶は得られなかった。溶媒の組み合わせを変更して検討を継続中である。また、推定構造をもつ化合物の有機合成を試みた。青色発色団の合成には成功したが、ペプチド鎖の脱保護の段階で基質が分解する結果となった。現在、保護基を変えて再検討を進めている。 Shellfish have shells in a variety of colors, but their pigments have yet to be chemically identified, and most of their biosynthetic pathways, roles, and evolutionary backgrounds remain rudimentary. In this study, we focused on the blue pigment contained in the green mussel (Perna viridis). The blue pigment has been found to be a pigment peptide with phenoxazine as a chromophore; but its stereochemistry remains to be elucidated. In this study, the objective was to determine the complete structure of this compound. The green mussels were collected in Enoshima, and their pigments were extracted. The amount of the blue pigment in crude extract was quantified by HPLC; its content was lower than previously obtained extracts (approx. 20%). The color of shells varies among individuals, with some individuals having a bright green color and others having a dull green color. It was suspected that the amount of blue pigment varied among populations. Using samples collected at Enoshima, Kanazawa, and Fukaura ports, image analysis and blue pigment quantification were conducted to examine the relationship between the two parameters; RGB decomposition unveiled that the Enoshima group had a significantly higher percentage of red elements (R%) and fewer G% and B% elements. Conversely, the other groups had less R% and more G% and B% elements. The quantification of blue pigment demonstrated that the Enoshima group had a lower amount, approx. 10-40% of that of the other groups. Blue pigment yield showed a negative correlation with R% and a positive correlation with B%. In appearance, the extraction from with bluer and less reddish coloration was found to yield more blue pigment. The extract was purified and approx. 80 μg of blue pigment was isolated; for structure determination by X-ray crystallography, the vapor diffusion crystallization of the pigment was explored. Thin and granular precipitates were obtained, but no crystals were done. The organic synthesis of the pigment with the presumed structure was also attempted; the blue chromophore was successfully synthesized, but the substrate degraded during the deprotection of the peptide chain.
