細胞はイオンチャネルを通る選択的イオン透過を利用して電気シグナルを生成する。そのため、イオンチャネルを通るイオンの選択性と透過は生命にとって必須である。本研究では、このイオン選択性とイオン透過の関係を調べることを目的とした。Punch through法による電流計測は、イオン種によって電流－電圧曲線の線形が異なることを示唆している。この違いを説明するため、分子動力学シミュレーションにより、モデルチャネルを通る電流－電圧曲線の線形とイオン透過の律速段階の関係について解明した。カリウムチャネルを通るナトリウムの電流－電圧曲線は優線形と予想され、これは低電位でのカリウム選択性に寄与すると考えられる。
Ion selectivity and ion transports through ion channels are essential for life: Living cells generate electrical signals by utilizing selective ion permeation through channels. The aim of this study is to reveal the relationship between ion selectivity and ion permeation. The electrophysiological measurement employing the punch through method implies that the shape of the current-voltage (i-V) curve differs among ion species. In order to explain this difference, the relationship between the shape of i-V curve and the rate-determining step of ion permeation was analyzed by using the molecular dynamics simulation. The shape of the i-V curve of sodium ions through the potassium channel was predicted to be superlinear, which is considered to contribute to the selectivity for potassium over sodium at low voltage.
