交雑系(129XC57BL/6)のGalT-I KOマウスは正常に発生するが,C57BL/6に8世代戻し交配したKOマウスは胎生後期から成長の遅延が認められ,出生時に100%死亡することを明らかにした。そこでさらにBALB/cに戻し交配すると,今度は胎生中期に致死となることがわかった。胎生10.5日ですでに発生の異常が認められ,出生前には胎児は吸収されてしまって存在しない。現在,その死亡原因を解析しているところである。また,マウスの遺伝的背景によってこのように大きく死亡時期が異なる理由についても今後解析していく。GalT-Iは主としてN型糖鎖の合成に関わると言われていたが,GalT-I KOマウス(交雑系)の赤血球膜上の糖鎖構造を詳細に解析したところ,Core2のO型糖鎖の合成も著しく阻害されていることが明らかとなった。Core2 O型糖鎖のβ-1,4ガラクトース側鎖はセレクチンのリガンド糖鎖であるシアリルLe^xの形成に必須であるので,このマウスはセレクチンのリガンド糖鎖の発現が低下している可能性が考えられた。白血球膜上のCore2のO型糖鎖の合成も著しく阻害されており,実際,このマウスの好中球や単球へのP-セレクチンの結合が低下していた。遅延型接触過敏症反応はセレクチンが重要な働きをしていることが知られているが,TNCBに対するこの反応がGalT-I KOマウスでは有意に抑制されていることがわかった。さらにその際,ランゲルハンス細胞のリンパ節への遊走能が阻害されていることを示す結果も得ている。以上の結果から,いくつか見つかったGalT遺伝子の中でもGalT-Iはセレクチンのリガンド糖鎖の合成に大きく関与しており,GalT-I KOマウスはセレクチンシステムの欠損による免疫系の異常が見られた。
