Note (General): 骨粗鬆症は骨折のリスク因子であり、生活の質の低下に直結する疾患である。ヒトゲノムワイド関連解析により、membrane palmitoylated protein 7(MPP7)遺伝子が骨密度に関与することが示された。その際、MPP7の発現低下が骨密度の低下と相関した。申請者らは、Mpp7遺伝子の欠損マウスを樹立し、生体内におけるMPP7の機能を解析した。MPP7欠損マウスは野生型マウスと同頻度で生まれた。雌のMPP7欠損マウスは野生型マウスよりも低い体重を示した。この体重の差は、雄のマウスでは観察されなかった。体重を除いては、Mpp7-KOマウスに特異的な異常は同定できなかった。ヒト細胞株を用いて、MPP7の蛋白の発現を調べたところ、MPP7は複数の骨肉腫由来の細胞株に発現していることが示された。以上の結果は、MPP7が生体内において体重の制御に関与すること、および樹立したMPP7欠損マウスがMPP7による骨密度の制御機構を解析する上で、有用なモデル動物になることを示した。
