Tokai High Avoider (THA) rat is a unique strain that maintains an inborn high learning ability. THA rats were developed from Wistar rats by a selective breeding of brother-sister rats showing high-avoidance rate in the lever-press Sidman avoidance test. To elucidate the molecular mechanism of higher brain function, we conducted a comprehensive metabolome analysis of Wistar rats (n=8) and THA rats (n=8), using capillary electrophoresis-mass spectrometry. As a result, we successfully identified 412 metabolites in the plasma, hippocampus, and cerebral cortex. Of the unique metabolic features elucidated in THA rats, we found significantly high concentrations of plasma branched chain amino acids (BCAA). These findings, in addition to elevated concentrations of BCAA and major components of the tricarboxylic acid cycle in the hippocampus, suggest that BCAA may play essential roles in THA rats, serving as an alternative energy source during exercise and possibly preventing the brain and muscle from central fatigue. Furthermore, differences in the glutathione pathway, urea cycle, and purine, pyrimidine metabolism suggested functional roles. Although biochemical verifications are needed, this study serves as an initial step in analyzing the association between metabolism and brain function by the use of metabolomics.
