Approach to the mechanism that only humans develop Parkinson's disease

パーキンソン病の代表的な病理として、α-Synucleinタンパク質の凝集が挙げられる。実際に、家族性パーキンソン病患者ではα-Synuclein の遺伝子変異が高頻度に検出されるため、強いリスク因子として考えられている。同一のリスク変異をヒト以外の大多数の哺乳動物がゲノム中に有しており、なお且つ、それらの動物種では凝集体(Lewy小体)の出現およびパーキンソン病の自然発症は認めない。上記の現象に基づいて、ヒト以外の変異を持つ動物種ではα-Synuclein凝集を「抑制」する機構が存在するという仮説を立て、本研究課題を計画した。
One of the most common pathologies of Parkinson's disease is the aggregation of alpha-synuclein protein. In fact, genetic mutations in alpha-synuclein are frequently detected in familial Parkinson's disease patients and are considered to be a strong risk factor. The same risk mutations are present in the genomes of most non-human mammals, despite the absence of aggregates (Lewy bodies) and spontaneous onset of Parkinson's disease in these species. Based on the above phenomenon, we hypothesized that there is a mechanism that "suppresses" α-synuclein aggregation in non-human species with mutations, and planned this research project.
