ミネラルコルチコイド受容体(MR)はアルドステロンをリガンドとする核内受容体型転写因子であり、高血圧や心臓、腎臓、血管の臓器障害に関わることが報告されているが、MR活性化機構の詳細は不明のままであった。我々は生化学的手法を用いて新規MR転写共役因子C14orf43を同定した。そこで、C14orf43の機能解析を通じてMR活性化の分子メカニズムを明らかにすることを本研究の目的とした。In vitroの研究により、CASZ1はMRとアルドステロン依存性に相互作用することが判明した。RNAiによってC14orf43の発現をノックダウンさせると、MRの標的遺伝子の1つであるSGK1のmRNA発現量が増加したことから、C14orf43はMRの転写活性を抑制する転写抑制因子であることが判明した。またアルドステロン処置によりMRやC14orf43はMR標的遺伝子ENaCαのプロモータにリクルートされることがChIPアッセイから確認できた。一方で、C14orf43自体には、転写を直接制御できるHDAC活性やクロマチンリモデリング活性を有するドメインを有さなかった。このことからC14orf43の機能として、MRと何らかの転写抑制性蛋白質複合体とのアダプターとして機能していることが予想された。そこで、C14orf43と相互作用する蛋白質複合体を生成するために、FLAGタグ付きC14orf43安定発現細胞株をレトロウイルスを用いて樹立した。この安定発現細胞株を大量培養し、核抽出液を取得後、FLAG抗体による免疫沈降を行い、C14orf43相互作用因子の精製を行い、得られたサンプルをLS-MS/MS質量分析を用いて同定したところ、HDAC1、HDAC2、CHD4、RBBP4、MTA2、MBD4など多くのエピゲノム制御因子が同定された。以上のデータから、アルドステロンによるMR活性化のメカニズムにおいてC14orf43がエピゲノムを介して転写を抑制していることが示唆された。
Mineralocorticoid receptor (MR) belongs to the nuclear hormone receptor superfamily and serves as an aldosterone-induced transcriptional factor. Although MR has been reported to be involved in hypertension and organ damage in the heart, kidney, and blood vessels, details of the MR activation mechanism remain unknown. We identified novel MR co-regulators C14orf43 by a biochemical approach. Therefore, we aimed to clarify the molecular mechanism of MR activation through functional analysis of C14orf43. In vitro studies revealed that CASZ1 interacts with MR in an aldosterone-dependent manner. Knockdown of C14orf43 expression by RNAi increased the mRNA level of SGK1, which is one of the MR target genes, and found that C14orf43 is a transcriptional repressor that suppresses the transcriptional activity of MR. It was also confirmed by ChIP assay that MR and C14orf43 were recruited to the promoter of the MR target gene ENaCα in an aldosterone-dependent manner. On the other hand, C14orf43 itself do not have a domain having HDAC activity or chromatin remodeling activity that can directly control transcription. So it is predicted that C14orf43 functions as an adapter between MR and some transcription repressible protein complex. Therefore, in order to identify a protein complex that interacts with C14orf43, we generated a FLAG-tagged C14orf43 stable expression cell line using a retrovirus. This stable expression cell line was cultured in a large amount, nuclear extract was obtained, immunoprecipitation was performed with FLAG antibody, C14orf43 interaction factor was purified, and the obtained sample was identified using LS-MS / MS mass spectrometry. As a result, many epigenomic regulators such as HDAC1, HDAC2, CHD4, RBBP4, MTA2, and MBD4 were identified. These data suggest that C14orf43 represses transcription via the epigenome in aldosterone induced MR activation.
