Elucidation of the molecular mechanism underlying anti-inflammatory activity of oxidized phospholipids in murine peritoneal macrophages

本研究は、酵素活性依存的に生成した酸化リン脂質に着目し、酸化リン脂質の炎症制御機構および責任分子の同定を目指して遂行した。マウス腹腔常在マクロファージは酸化リン脂質合成酵素12/15-lipoxygenase(12/15-LOX)を高発現することが知られている。実際に申請者はこれまでに12/15-LOX欠損マクロファージにおいて、12-HETEや14-HDoHEをアシル基に有する酸化リン脂質が顕著に減少していることを見出している。そこでまず、野生型および12/15-LOX欠損マウスから単離した腹腔常在マクロファージの炎症応答を比較することで、酸化リン脂質の炎症応答に対する機能を検証した。野生型および12/15-LOX欠損マクロファージをLPSで刺激し、炎症性サイトカインのmRNA発現量をリアルタイムPCRで解析した。その結果、野生型に比べて12/15-LOX欠損マクロファージでは、IL-6やIL-1bなどの炎症性サイトカインのmRNA発現量が顕著に亢進した。また、12/15-LOX欠損マクロファージに各種酸化脂肪酸を添加した結果、DHA由来代謝物である14-HDoHE添加により炎症応答の抑制傾向が見られた。以上の結果から、14-HDoHEが膜リン脂質に取り込まれ、酸化リン脂質となることで炎症を制御する可能性が考えられた。今後は、14-HDoHEが実際に膜リン脂質に取り込まれているか、ならびに取り込みを阻害した場合に抗炎症作用が消失するかを検証する必要がある。
In this study, we aimed to characterize the role of oxidized phospholipids (OxPLs) on inflammation. Murine peritoneal macrophages highly express 12/15-lipoxygenase (12/15-LOX) which can directly oxidize polyunsatulated fatty acyl chain in phospholipids and generate OxPLs in a 12/15-LOX-dependent manner. We previously reported that several molecular species of OxPLs were significantly lower in 12/15-LOX knockout (12/15-LOX KO) peritoneal macrophages than wild-type (WT) peritoneal macrophages.
At first, we compared the mRNA expression levels of pro-inflammatory cytokines such as IL-6, TNF-a and IL-1b in 12/15-LOX KO macrophages with WT macrophages. As a result, the mRNA levels were significantly higher in 12/15-LOX KO macrophages than in WT macrophages, suggesting that 12/15-LOX-derived OxPLs have anti-inflammatory activity in murine peritoneal macrophages. We next exogenously added a series of free oxidized fatty acids to peritoneal macrophages isolated from 12/15-LOX KO mice and measured the cytokine mRNA levels. In 14-HDoHE-treated macrophages, the mRNA levels were tend to be lower than other oxidized fatty acid-treated macrophages. 14-HDoHE might exert anti-inflammatory effect by being incorporated into cellular phospholipids.
