Clarification of neural circuit development towards the understanding of developmental disorders

発達障害の病態には様々な遺伝的要因と環境要因が関与する。一方で、発達障害には共通して、社会性障害や限定的な興味など、特定の症状が見られることから、発症に至る共通のメカニズムが存在すると考えられる。本研究では、発達障害の背景にある「共通原理」の解明を目指し、病態の根幹と考えられる神経回路網の成熟過程を明らかにすることを目的とした。回路成熟機構の分子基盤の解明のためにはシナプスオーガナイザーであるC1qファミリー分子の機能を追究している(Ito-Ishida, Neuron, 2012; EJN 2014)。また、病態モデルマウスの前頭前野機能を解析するために新たな行動実験系の開発をすすめた。さらにモデルマウスとしてMecp2欠損マウス(Ito-Ishida, Nature Neuroscience, 2018 およびJNS 2020)を用い開発した手法を用いて回路異常の実態を明らかにしている。
Multiple factors concerning genes and environments lead to developmental disorders such as autism. While the exact pathophysiology remains elusive, the existence of features observed across patients with these disorders suggests that there must be a common pathway leading to the neurological deficit. One of the features of particular interest is the altered long-range connectivity, which has been reported by studies using fMRI. We have thus aimed to clarify the molecular mechanism regulating neuronal connectivity by focusing on the roles of C1q family proteins, which work to establish synaptic connections between different brain regions (Ito-Ishida, Neuron, 2012; EHN, 2014). In addition, to achieve efficient behavioral analysis in mouse models, we have established a novel automatic behavior platform, which enables analysis of the prefrontal cortex-dependent tasks in mice. We are currently analyzing Mecp2-related mouse models (Ito-Ishida, Nat Neuro, 2018; JNS, 2021) to detect circuit-wide abnormality and its consequences.
