Recently, Naito et al. (1999) showed that latencies of wave I of ABRs were significantly prolonged in central myelin deficient hamsters. They suggested that central myelin deficiency leads peripheral auditory dysfunction. In this study, we examined the ABRs, DPOAEs and cochlea morphology of myelin deficient (md) rat, which completely lacks central myelin but not peripheral myelin. ABRs showed a marked prolongation only in wave I latency recorded at lower tone-burst stimulation. Demylinated CNS tissue protrusion was observed in the center of the cochlear nerve in the internal auditory canal. DPOAEs decrease at lower frequency of the combined tone. Since nerve fibers ending at the apical turn of the cochlea passed through central portion of the cochlear nerve, wave I prolongation of ABRs and decrease of DPOAEs at lower frequency might originate mainly in demyelinated CNS part of cochlear nerve in the internal auditory caval.
