ペクチンは複数の構成糖から成る植物細胞壁多糖の一つであり、その主要な構造は、ガラクツロン酸(GalA)が α-1,4-結合で連なったポリガラクツロン酸(polyGalA)である。polyGalAはラムノース残基を介して結び付けられており、ラムノースの一部には中性糖鎖が分岐結合している。また、GalAの一部はメチル基やアセチル基で修飾されている。本研究は、ペクチンの合成機構に着目し、 polyGalAにGalAを転移させて糖鎖の伸長反応を触媒するガラクツロン酸転移酵素(GTase)の諸性質を明らかにすることを目的とする。1. 酵素の可溶化:アズキ(Vigna angularis)を室温、暗所、7日間生育し、胚軸から粗膜画分を調製して酵素源とした。GTaseの活性測定方法は確立し、また、酵素の諸性質も明らかにした。本年度は酵素の可溶化を検討し、可溶化酵素液からの酵素の精製を試みた。膜画分を終濃度が50mM Tris-HCl緩衝液(pH7.5)、1%TritonX-100、20%グリセロール、1mM EGTA溶液と混合し、時々撹拌しながら氷上で60分間放置した。粗膜画分の約90%の活性が可溶化された。可溶化された粗酵素液を、DEAE-セルロースカラムを用いたイオン交換クロマトグラフィー、アフィニティークロマトグラフィー、ならびにNiキレートカラムを用いたクロマトグラフィーも試みたが良好な結果は得られなかった。2.GTaseの活性測定にはUDP-GalAが基質となる。本化合物は市販されていないので、有機化学的手法を用いて合成を行った。ガラクトースペンタアセテートを出発材料とし、15段階の反応によりガラクツロン酸1-リン酸を合成した。本化合物をUMP誘導体とカップリングし、UDP-GalAを合成した。
