嫌気性セルロース分解性細菌Clostridium cellulovorans の生産するセルロソームと呼ばれる酵素複合体は、セルロースやヘミセルロースを効率的に分解する。セルロソームは、酵素活性を示さない骨格タンパク質(CbpA)とセルロソーマル酵素群の2 種類のコンポーネントからなる。CbpA は、セルロース結合ドメイン(CBD)、Cohesin ドメイン、SLH(Surface Layer Homology)ドメインを持つ。セルロソーマル酵素はCbpAのCohesinドメインに結合するためのDockerinドメインを持ち、CbpAと酵素複合体を形成し、セルロースを効率的に分解できる。C. cellulovorans では、セルロソームのほかにセルラーゼや他のコンポーネントが協調的に働くことでセルロース分解における高い活性を示す。しかしながら、嫌気性菌であるC. cellulovorans は培養が難しく、また遺伝子組み換え技術が確立されていないためセルロソームの研究・利用は容易ではない。そこで、大腸菌とともに遺伝子組み換え技術で実績のある枯草菌を用いて、セルロソームの解析・改変を行った。
