豆乳を始めとした大豆加工食品の消費量は年々増加傾向にあり、安全安心な製品を供給することは食品メーカーの責務である。そのため、最終製品の微生物検査は信頼性が高く簡便で迅速であることが望まれる。標的の微生物種をコロニーとして形成させる従来の培養法は培地調製の手間がかかる、経験が必要や培養時間が長いなどの様々な問題点がある。近年、多くの微生物迅速評価法が開発されているが、食品成分による阻害が導入の障壁となっている。そこで本研究では大豆加工食品中の微生物数及び微生物種を正確に迅速に評価できる手法としてフィルター染色法による迅速計測、MALDI-TOF MSを用いた直接同定、自動生菌数測定装置の導入を検討した。その結果、等電点沈殿による除蛋白や界面活性剤による油分除去といった適切な前処理手法を開発することで、迅速計測、迅速同定及び自動計測法を導入することが可能となった。食品事業者にとって微生物検査の結果を迅速に判断できるということは微生物管理システムの高度化を意味し、より安全な製品作りに注力可能となる。本研究は大豆加工製品の安全、安心の保証に高く貢献出来るものと考えられる。
