ワクチンが有効に働く為には抗体遺伝子の体細胞突然変異が必須である。遺伝子変異はまた我々の体細胞にとってはがんを発症する危険因子となる。B 細胞には体細胞遺伝子変異を高率に誘導するとともにそれに対する安全性を確保する機能が必要である。本研究は、末梢リンパ組織のB 細胞で発現が上昇するGANP 分子が抗体の体細胞遺伝子変異を安全に行わせることを明らかにした。動物実験においてGANP の機能破綻が、がん発症を来たすことを証明し、また臨床検体によってもリンパ腫、脳腫瘍、メラノーマ、胆管がん、乳がん等のがん化と関連する可能性を示した。本成果は自然発がん、炎症発がんの診断、治療に寄与する。
Antibody response to T cell dependent antigens accompaniessomatic hypermutation of immunoglobulin Variable region genes, which needs to beregulated precisely and safely. We studied the role of a molecule named GANP up-regualtedin germinal center B cells in the peripheral lymphoid organs. GANP is essential foraffinity maturation of the antigen specific B cells. Impairment of GANP function causedlymphomas and mammary gland tumors in mice. We showed that the GANP impairments areassociated with tumor development in humans and provided the information regardingspontaneous cancer development during inflammation.
