マウス骨髄ストローマ細胞にINK4a-/-、c-Myc過剰発現の条件を付加することにより骨肉腫の発生が確認された。single cell cloningにより骨肉腫のtumor initiating cellとして骨、軟骨前駆細胞(AX細胞)と間葉系幹細胞(MSC)の性質を有する(AO細胞)2種の細胞が同定された。前者の分画は発癌能が高く骨肉腫幹細胞の性質を有していた。後者の細胞からの発癌には脂肪分化能の喪失が必要であった。また骨肉腫幹細胞にPPARγを導入し脂肪分化能の回復させたところ腫瘍形成能が抑制された。以上から脂肪分化は骨肉腫形成に負に働くことが解明され、発癌には分化の因子も重要であることが明らかとなった。
