中心体は母・娘中心小体と中心体周辺物質から構成されるオルガネラであり、1細胞周期あたり1回のみ複製されるように厳密にコントロールされている。中心体複製のコントロールが外れ、ゲノムDNAの複製周期と中心体の複製周期が乖離すると、多極分裂や細胞質分裂異常が生じ、複製したゲノムを正確に娘細胞に分配することができなくなり、癌細胞の発生または悪性化を招くと考えられている。今回、娘中心小体を特異的に染色する抗体を作成し、ヒトボーエン病臨床検体において、母・娘中心小体を別々に染色することに成功した。結果、ボーエン病組織において中心体複製異常が存在することが明らかとなった。母・娘中心小体がそれぞれほぼ同数増えていたことから、ボーエン病組織における中心体複製異常が細胞質分裂の失敗により起こっていることが示唆された。さらに、ボーエン病モデルマウスとして、中心体制御キナーゼであるAurora Aの皮膚特異的過剰発現マウスを作成した。本欠損マウスでは、基底細胞層に多核の細胞を生じ、その多核細胞では中心体の複製異常が観察され、ボーエン病の組織学的異常を反映したモデルマウスであると考えられた。
