慶應義塾大学病院で行われた生体肝移植ドナー手術において、術後1週間・1カ月後の肝再生能力を年齢別に検討した。ドナー手術は、ヒトにおける正常肝の肝再生が観察可能な唯一の臨床モデルであり、若年ドナー(30歳未満)の1週間後の残肝肥大率が、高齢ドナー(50歳以上)に比して有意に高いとの結果を得た。生体肝移植ドナー肝の生検検体または、切除された転移性肝腫瘍の正常肝組織の一部を患者の同意のもとに採取し、ヒト正常肝の幹細胞マーカーであるThy-1を使って肝幹細胞の分離を試みたところ、高齢になるにつれてThy-1陽性細胞のpopulationが低下する傾向を認めた。本研究によりヒトの肝再生能力が加齢に伴い有意に低下する事が示され、そのメカニズムにThy-1陽性細胞が関与している可能性が示唆された。
