われわれはニコチンが成長軟骨細胞に直接かつ特異的に作用して内軟骨性骨化に影響を与えると推測し、本研究を行なった。成長軟骨細胞3次元培養において、ニコチンは容量依存性に軟骨基質産生と肥大化を抑制し、同細胞においてalpha7 nicotinic acetylcholine receptorの発現を認めた。ニコチンは野生型マウス胎児の骨格成長を抑制した。ニコチンは軟骨細胞の特異的受容体を介し、基質産生と肥大化を抑制し内軟骨性骨化を遅延させると推測された。
