低酸素培養は輪部より分離したヒト角膜上皮幹細胞の増殖を促進する。酸素濃度はHypoxia inducible factor(HIF)によって感知され、HIF の酸素感受性のalphaサブユニットは3種類知られる。本研究では、ヒト角膜上皮幹細胞におけるHIF-aの発現と機能を解析した。RT-PCR解析によれば、初代培養ヒト角膜輪部上皮細胞及びヒト角膜上皮細胞株はHIF-1a, -2a, -3aを発現していた。siRNAを用いた抑制実験によれば、HIF-2aの抑制により増殖が抑制されたが、HIF-1aの抑制は細胞増殖に影響を与えなかった。Flow cytometry解析によれば、HIF-2a抑制群ではG2期にある細胞が増加していた。なお、HIF-2aの過剰発現は増殖に影響を与えなかった。これらの結果はHIF-2aと角膜輪部上皮細胞の増殖との関連性を示唆する。
