ADAM28の癌細胞転移への関与を調べるために、ルシフェラーゼとVenus融合タンパク質(ffLuc-cp156)を恒常的に発現する肺癌と乳癌細胞株(PC-9ffluc-cp156とMDA-MB231ffluc-cp156)を作製し、マウスの尾静脈内注入による肺転移モデルと乳房皮下脂肪組織移植による自然転移モデルを用いて癌転移を解析した。バイオイメージングを用いた解析法によりPC-9ffluc-cp156細胞の肺転移は、shRNAによるADAM28の発現抑制とADAM28に対する中和抗体処理により有意に抑制された。また、MDA-MB231ffluc-cp156細胞でもshRNAによるADAM28発現抑制で局所での癌細胞の増殖が抑制され,ffLuc-cp156に対するプライマーを用いたRT-PCRの解析から、各臓器(肺、心臓、肝臓、腎臓、脳など)への転移が有意に抑制された。さらに、PC-9ffluc細胞のマウス尾静脈内注入後におけるマウス血漿中VWFの分解は、mock群に比べてshADAM28群で抑制されていた。以上のことから、癌細胞に発現するADAM28はVWFを血中で分解し、VWF誘導性アポトーシスの回避により癌細胞転移促進に働く可能性が示唆された。
