我々は新規に樹立した骨肉腫マウスモデルを用いて、非腫瘍細胞(線維芽細胞、マクロファージ)から産生されるFgf2、Lifなど液性因子がErk1/2を強力に活性化し、骨肉腫細胞の未分化維持に関わることを明らかにした。さらにFgf2 は骨肉腫細胞の増殖、遊走を促進し抗癌剤への感受性低下にも関与した。Fgfr阻害剤を用いたin vivoの治療効果検証ではアドリアマイシンの効果を増強させた。以上の結果は、これらの液性因子が分化・非分化部を有する骨肉腫の病理・病態像形成に深く関わること、下流のシグナル伝達経路が骨肉腫の病勢を抑える有望な治療標的となる可能性を示唆する。
