本研究は、癌幹細胞の主要なマーカーであるCD44によって制御される糖代謝シグナルが癌治療の新たなターゲットとなりえるかについて検討することを目的として研究を行った。まず、CD44の細胞内領域とPKM2の相互作用が癌細胞の糖代謝に関与することが明らかになったため、グルコース代謝の重要なステップを触媒する酵素pyruvate kinase M2の活性に関わるチロシンリン酸化ステータスの変化を検討した。その結果、CD44の発現抑制はPKM2のチロシンリン酸化状態を低下させるとともに、その活性を負に制御することで、癌細胞の好気性解糖経路を促進することが分かった。以上のようにCD44を介した癌細胞の糖代謝制御が、癌治療のための新たな分子標的となり得ることが明らかとなった。
