抗加齢遺伝子Sirt1の腎臓での作用のうち、特に近位尿細管(PT) Sirt1に注目し、検討を進めている。我々はナトリウム・リン共輸送体(Npt2)promoterを使用し、近位尿細管特異的Sirt1過剰発現マウス(Tg群)を作成し、これらのマウス(control= WT, Tg)を用い、CKDの内、特に、糖尿病性腎症(DN, Diabetic Nephropathy)の解析を進めている。DNの発症初期においては(Control マウス)、まず近位尿細管のSirt1が低下する。その後、Sirt1低下がpodocyteに波及し、podocyteにおけるSirt1の発現も低下する。その結果podocyteのスリット膜形成分子が低下する。これにより、足突起の癒合・アルブミン尿が生じる。(これらの病態はSirt1Tgで抑制を認めた)。現在、Sirt1低下によるpodocyte機能異常のメカニズムを解明するため、更に詳細な検討を進めている。
