本研究課題名にあるTREM1のsheddingに関しては, 生体内で観察した結果を生化学的手法にて再現できなかったため, 対象遺伝子をIL-1受容体に変更し, 立案計画に沿って解析を続行した. IL-1は炎症および感染防御において重要な機能を有するサイトカインであり, その活性は極めて高度に調節されている. IL-1の受容体には2種(IL-1RI, IL-1RII)存在するが, IL-1RIIは細胞内領域を欠損しており, IL-1シグナルを負に調節すると考えられている. さらに, これらの受容体は細胞膜上にて蛋白質分解作用(以下, shedding)をうけ, 可溶型蛋白質としても存在することが報告されている. 昨年度は, 各種蛋白質分解酵素阻害剤, TACE遺伝子改変マウスおよびTACE欠損細胞を用いて生化学的な評価を試みた. その結果, IL-1受容体のうちIL-1RIIのみがTACEにて選択的にsheddingを受け, TACEはIL-1RIIのsheddingを介して細胞内のIL-1シグナルを正に制御していることが示された. 本年度は, TACE遺伝子改変マウスの骨髄細胞を用いてFACS解析を行い, IL-1RIIのみがTACEにて選択的にsheddingを受けることを確証, さらにIL-1RII変異体を作成しIL-1RIIは細胞膜近傍で切断されることを確認した. またヒト細胞やTACE欠損細胞を用いてIL-1シグナル解析を行った. 炎症性疾患・免疫に極めて深く関与しているIL-1シグナルの調節機構を解明することは, 臨床応用へのフィードバックの一助となると考える.
