細胞の代謝は転写、翻訳、酵素反応のレベルで制御されており、これらの複数の階層に跨ってどのように代謝が調節されているのかは大変興味深い。私は大腸菌の野生株や⊿arcA株(硝酸条件)を複数の培養条件で連続培養し、トランスクリプトーム、メタボローム、代謝フラックスを測定し、それぞれの調節機構を明らかにした。また、測定したマルチオミックスデータを構築した動的モデルを使って統合することで動的モデルのチューニングを実施し、⊿arcA株の遺伝子発現比を元にシミュレーションしたところ、予測された反応速度は実測フラックスとほぼ一致することを示した。異なる階層のオミックスデータを統合し、細胞内の調節機構を解明するためのプラットフォームとしての有用性を示すことができた。
