本研究では、人工多能性幹細胞 (iPSC)の初期化因子であるLin28、Nanog、Klf4の新たな機能を明らかにするために、それぞれに関して我々が開発した独自技術のin vitro virus (IVV) 法を用いてタンパク質間相互作用の解析を行った。その結果、Lin28の相互作用因子としてTnp2が、Nanogの相互作用因子としてNap1が、Klf4の相互作用因子としてegfr mRNAがそれぞれ同定された。Tnp2 とLin28は、Msi1を強制発現させたmESCにおいて共局在し、相互作用することが明らかになった。Nap1をノックダウンすると、Nanog、Oct4、Sox2の発現量が減少し、過剰発現させるとこれらの初期化因子のmRNA の量が増加した。その結果、Nap1はNanogと相互作用し、その転写活性を促進していることがわかった。細胞増殖およびアポトーシス抑制に関わるegfr mRNAのCDS領域断片が配列特異的にKlf4 と結合することが示された。さらに、ヒト前立腺癌細胞PC-3にKlf4を一過性発現させた結果、EGFRタンパク質の大幅な発現量上昇が確認できた。マウス線維芽細胞からiPS細胞誘導までのEGFRおよびKlf4の発現量推移を経時的に観察した結果、双方の発現量上昇が同じ段階で起きていることから、iPS細胞誘導時においてもKlf4がEGFRの発現量を上昇させている可能性が示唆された。
