超好熱性アーキアであるPyrococcus furiosusをモデル生物として用い、同アーキア由来でRNA鎖の連結に関わると考えられる3種の酵素に対応する遺伝子を単離し、その組換え体タンパク質を大腸菌にて産生、精製した。これらは、我々が発現クローニング法にて見いだした(a)2'-5' RNA リガーゼ様のPF0027タンパク質。また既に報告がある酵素のホモログとして同定した(b) T4 RNA リガーゼタイプのPF0353タンパク質、及び(c) RtcB リガーゼタイプのPF1615タンパク質である。さらに、これらの酵素と関連する複数の遺伝子を単離し、その組換え体タンパク質を用いて試験管内のRNA 連結反応を解析する系を構築した。
