in silico解析により着床前期胚特異的新規遺伝子を抽出し、ノックアウトマウス(KO)を作成し機能解析を行った。Kzpi(KRAB-zinc finger)のKOは成獣まで発生し、生殖能も維持されていたが、有意な産仔数の減少、早期死亡傾向、腟閉鎖などの異常が認められた。Kzpi 欠失ES 細胞では異常な表現型は認められなかったが、インプリンティング遺伝子の発現異常、アレル特異的メチル化領域(differentially methylatedregion: DMR)の低メチル化が認められた。Kzpi は、初期胚でのゲノムワイドな脱メチル化から、DMR 特異的なメチル化を維持すると考えられた。Zfpi(SCAN-zinc finger) KO は成獣まで発生し、生殖能も維持されていたが、脾細胞の核型解析Q-band で多数の不完全断裂部位が認められた。Zfpi 欠失ES 細胞についても解析中である。
