血液のがんである骨髄腫は難治性であり、生命予後は悪い。本研究では先天的にリンパ球と補体を欠損する重度免疫不全マウスを用いてヒト骨髄腫細胞の挙動と病態の解析を行い、抗がん剤治療モデルを作成した。マウスに移植されたヒト骨髄腫細胞は血流が豊富な骨幹端に集まり、局所では骨芽細胞、破骨細胞と接触していた。また、骨髄腫細胞の増殖に伴い骨破壊が進み、病的骨折を合併した。移植されたがん細胞の一部は低酸素状態において接着分子カドヘリンを発現し、抗がん剤に対する薬剤耐性を獲得した。本研究により骨髄腫の骨微小環境における動態ならびに抗がん剤の耐性獲得の機序があきらかとなり、新薬開発につながると期待された。
