本研究では、上皮性卵巣癌の標準化学療法であるTC療法(CBDCA+PTX)の効果を予測できる遺伝子を決定した。上皮性卵巣癌で初回手術時に評価可能病変を認めた症例で、TC療法の効果を予測できる遺伝子をスクリーニングし、そのスクリーニングした遺伝子のうち155遺伝子によりsiRNAの設計をしトランスフェクションプレートを作成した。トランスフェクションプレート上で、TU-OS-4細胞を10000個を播種し、CDDP、PTX濃度を各13uM、45nMで48時間暴露してCell Counting Kit-8にてIC50を算出した。そしてTC療法細胞死阻害遺伝子およびTC療法細胞死誘導遺伝子を選別した。さらにトランスフェクションプレートを構成した155個の遺伝子により、カスタムアレイ(発現解析)を作成した。上皮性卵巣癌で初回手術時に評価可能病変を認めたTC療法を施行しCRであった症例6例NC/PDであった症例7例を新たに集積し、作成したカスタムアレイを施行し、各遺伝子の効果を予測できる精度を算出した。TC療法細胞死阻害遺伝子およびTC療法細胞死誘導遺伝子を選別した。TC療法でCRかNC/PDを判別できる精度を、各遺伝子毎にROC曲線を作成して検証したところ15遺伝子でROC曲線のAUC0.7となった。
