エピゲノム情報はその分化可能性を評価する指標となることが考えられるが、どのような領域のエピジェネティック状態が細胞の性質に影響しうるかは明らかでない。本研究では、細胞種に特徴的な未知のエピゲノム変化を探索し、幹細胞の分化可能性評価のためのエピゲノム情報基盤を整備することが目的である。本研究計画内において、申請者らは心筋細胞特異的なgene body領域の低メチル化状態を見出した。広範囲に渡る低メチル化状態は、転写およびエピジェネティック因子の蓄積を伴っており、細胞の特性に密接に関わるエピゲノムを形成する一因となる。本研究計画では、エピゲノム情報による幹細胞評価系の整備に重要な細胞特異的エピジェネティック状態を見出した。
