CD44は接着分子としてがん幹細胞の自己複製能を維持するために必要な因子の一つと考えられていることから人工がん幹細胞が細胞プレートへの接着を阻害する一本鎖抗体のセレクションをIVV法により行った。全部で７ラウンドの選択実験後、得られた2つのクローンAXT7-30とAXT7-12はAXT細胞の細胞プレートへの接着を阻害した。CytoTox-Glo Cytotoxicity Assayの結果、強い殺細胞活性を有し(IC50=3nM)、AXT7-30の方が3倍強い活性を有していた。そこでこの一本鎖抗体AXT7-30を使って蛍光ラベル化抗体を作成した。tdTomatoは非常に明るい赤色蛍光タンパク質であり、tdTomato蛍光ラベル化されたAXT7-30を用いて免疫染色したところAXT細胞を明瞭に蛍光観察することができた。現在この蛍光ラベル化一本鎖抗体の担がんマウス投与を実施中であり、二光子励起蛍光顕微鏡を用いた体内動態の画像化によりがん細胞への集積を解析中である。
