当研究では肺動脈性肺高血圧(PAH)の病態解明、新規治療法開発のため、BMPR2あるいはAlk1遺伝子変異を有する重症肺高血圧患者からiPS細胞を樹立し血管内皮細胞に分化誘導し機能解析を行った。低酸素化培養によって肺環境を模倣したところAlk1遺伝子変異を有する肺高血圧患者由来の血管内皮細胞(PAH-EC)では管腔形成能の亢進を認め、病態形成に関与していると考えた。またAlk1の下流であるSmad1,5、BMPR2の下流であるSmad2,3のリン酸化に関しては明らかな差異は認めず、未知のシグナル経路が病態に関与している可能性が考えられた。
