まずIn vitroにてマウス膀胱癌細胞株MBT-2variant細胞にリポソーム法にてLacZプラズミド導入を行い、X-gal染色にてプラズミドの効率的な導入を確認した。また同様にPTENプラズミド導入も行い、Western blot法にてMBT-2variant細胞内でのPTEN蛋白の発現の持続、Akt活性の低下が確認された。次にC3H/HeNマウスの膀胱内へMBT-2variant細胞(5×105乗cells)を注入することにより、マウス膀胱癌同所性モデルを作成する手技を確立し、このモデルでは30日目頃にマウスは癌死することがわかった。In situでの膀胱内へのLacZプラズミド導入では、特に腫瘍巣に強いX-gal染色を認め、効率的な遺伝子導入が insituでも行われていることが確認された。よって本手法は、筋層非浸潤性膀胱癌に対する膀胱内注入治療確立の布石となる可能性があると考えられた。
