間葉系幹細胞(MSCs)は主に骨髄に存在し、in vitroにおいて脂肪・軟骨・骨など間質細胞への分化能(多能性)と自己複製能を持つことが解析されてきたが、in vivoでの局在、表現型や発生学的起源は不明なままである。本研究では、近年我々が見出したSca-1およびPDGFRα表面抗原の発現を指標に胎生期MSCsを解析し、Sca-1の発現パターンが胎生(E)14.5-16.5日目から変化することを見出した。さらに細胞系譜を追跡するためにSca-1遺伝子座にCreERT2 2A KOrをknock inしたES細胞を作製し、germline キメラマウスを得た。
