作用点の異なる様々な小分子化合物によるUPR関連遺伝子群の発現パターンを比較解析した結果、ストレスセンサーの活性とUPR関連遺伝子の発現が化合物の作用点の違いによって少なくとも3パターン存在することが示唆された。また、小胞体ストレスセンサーの一つとして知られるIRE1αの活性化をイメージングするために、IRE1αの活性化に必要十分なステップである二量体化を、分断された蛍光蛋白質の再構成によりタンパク質間相互作用を検出する手法Bimolecular Fluorescence Complementation (BiFC)法を用いて検出する系を構築した。 In this study, we compared the expression patterns of nine UPR target genes induced by seven UPR-inducing compounds with different modes of action. As a result, it was suggested that the existence of at least three types of ER stress sensors' activation profiles and UPR target gene expression profiles, which depend on the mode of actions of the compounds. Moreover, we developed a new detection system for the activation of IRE1a by evaluating dimerization of it by bimolecular fluorescent complementation (BiFC) assay. By detecting the fluorescence derived from the reconstituted cerulean, this assay system enabled us to distinguish the activation behaviors of IRE1a as to ER stress-inducing compounds.
