高血圧の発症には遺伝素因と環境因子の双方が重要な役割を担うことが知られている。環境因子の中でも特に塩分が重要な役割を担うことが示されているが、塩分が高血圧の発症とどのように関連するのかについては不明な点が多い。本研究では2つの高血圧モデル動物に対して、6週齢から14週齢まで高食塩食を投与すると、高血圧が持続する、すなわち「塩分メモリー」が存在することを明らかとした。また、腎臓の交換移植実験を行った結果、その主座が腎臓に存在することを明らかにし、またその機序を検討した結果、腎臓の細動脈のリモデリングと全身および局所のレニンーアンジオテンシン系の亢進がこの現象に関与している可能性が示唆された。 Hypertension is known to occur because of an interplay between genetic and environmental factors. Of these environmental factors, changes in salt balance are thought to play a major role in the pathogenesis of hypertension, but the precise mechanisms by which the changes in salt balance cause elevations in blood pressure are still unclear. In this study, we show that transient treatment with a high-salt diet from age 6 to age 14 weeks causes hypertension which is sustained even after the high-salt diet is changed to a normal diet. Analysis of the mechanisms involved by renal cross-transplantation revealed an important role for the kidney in the salt memory phenomenon and also revealed that changes in renal vascular hypertrophy and a sustained increase in the systemic and local renin-angiotensin system were involved in the molecular mechanism.
