ゲノムへの遺伝子挿入が無いエピゾーマルプラスミドもしくはセンダイウイルスを用いてヒト線維芽細胞に山中4因子(Oct4, Sox2, Klf4, cMyc)の導入を行い、これまでのレトロウイルスを用いた方法よりも効率よくdiNSCを含むニューロスフェアが形成されたが、それらのスフェアを分化誘導したところ、神経分化が極めて遅いという問題が明らかになった。一方でiPS細胞の樹立は線維芽細胞から末梢血由来細胞に変化してきており、ヒトdiNSCも末梢血由来細胞から誘導できるかを検証したが、良好な神経分化を得られる誘導方法の確立には至らなかった。 We succeeded in making human directly induced neural stem cells by using Sendai virus vectors. Although we were able to obtain increased number of neurospheres compared to retroviral vectors, SeV-induced neurospheres did not differentiate into neural cells efficiently.
