ヒト心臓線維芽細胞および皮膚線維芽細胞を培養して心筋リプログラミングを行った。マウスで有効であった3つの遺伝子Gata4, Mef2c, Tbx5ののみではヒト細胞の心筋誘導に不十分であることが分かった。そこで新たに心筋細胞特異的に発現している11の遺伝子の中からヒト心筋リプログラミングに必須の因子をスクリーニングした。その結果、GMTにさらに2因子を加え5因子にすることでヒト心筋リプログラミング効率が改善することを見出した。5因子により誘導されたヒト心筋様細胞は心筋特異的な遺伝子発現を示し、また心筋に特徴的な生理機能も確認できた(Wada et al, PNAS, 2013)。 Direct reprogramming of differentiated human somatic cells into cardiac cells holds great promise for regenerative medicine. It has not been whether human fibroblasts can be directly reprogrammed into differentiated cardiomyocytes by defined factors. Here, we demonstrate that Gata4, Mef2c, and Tbx5 could also convert postnatal human cardiac fibroblasts directly into cardiomyocyte-like cells, when combined with two additional cardiac transcriptional regulators. Human induced cardiac cells expressed multiple cardiac-specific markers, changed a global gene expression profile toward a cardiomyocyte-like state, and generated spontaneous Ca transients. Importantly, a subset of induced cardiac cells matured to exhibit action potentials and contracted synchronously when co-cultured with primary murine cardiomyocytes. Thus, human fibroblasts can be converted into cardiomyocyte-like cells and these methods may facilitate future applications in regenerative purposes.
