本研究では、発達期小脳に豊富に存在するD-セリンの生理的役割について追究した。その結果、D-セリンは神経活動依存的にグリア細胞から放出され、神経細胞に発現するデルタ2型グルタミン酸受容体と結合することにより、記憶・学習の分子基盤とされるシナプス可塑性を制御することがわかった。興味深いことに、デルタ2受容体は細胞内最C末端領域で結合するチロシン脱リン酸化酵素PTPMEGを介してシナプス可塑性を調節していることが示唆された。したがって、本研究で見出された新規D-セリンシグナリングは、グリア－ニューロン相互作用の理解を深める上で有益な知見を与えうるものと期待される。 In this study, we examined the functional role of a D-serine highly existing in the developing cerebella. D-Serine in the cerebellum, which was released from glial cells in a neuronal activity-dependent manner, bound to the postsynaptically-expressing delta2-type ionotropic glutamate receptors (delta2 receptors) to regulate synaptic plasticity, a molecular basis of learning and memory. Interestingly, we revealed that delta2 receptor controlled the synaptic plasticity by binding to the tyrosine phosphatase PTPMEG via its intracellular C-terminus. Thus, D-serine-delta2 signaling is a unique D-serine signaling, which may gain new insights into the glia-neuron interaction.
