脱細胞化臓器骨格は、再生細胞の足場として有用であるだけでなく臓器特有の三次元構造から外科的に移植可能なマクロ構造へ連続する血管網、細胞外マトリックス構造を有している。我々はiPS細胞技術の臨床応用に向けて、ブタの肝臓を用いて脱細胞化骨格の移植手技の定型化に努めてきた。移植後の長期的機能維持を目指し、還流血液の凝固の問題をクリアするため、内皮細胞や抗凝固粒子による門脈系の被覆、門脈カテーテルを用いた抗血栓薬の持続投与によって大幅な改善を得た。今後は、膵臓・小腸を含めた本技術の幅広い応用の可能性を含め、脱細胞化臓器骨格を基盤としiPS細胞を用いた再生医療を実現すべく更に本研究を推進していきたい。 Recent technology in stem cell engineering has raised new possibilities for the study of complex physiological processes in vitro, as well as the potential to fabricate entire organs for transplantation. After the huge progress in vitro using human-induced pluripotent stem cells which can differentiate relatively functional somatic cells, we should go for the next stage to investigate clinical applications of those cells using feasible animal models. Therefore, we have been developed a new technology using acellular three-dimensional scaffolds to generate transplantable whole liver which can be recellularized with appropriate cell types. It can retain all the necessary cues of tissue regeneration with efficient blood supply by suturing vascular access in vivo. For the clinical application, it should be applied to preclinical experimental large animal models. We believe that this technology may help to open new insight into organ regeneration as well as drug metabolism and toxicity.
