本研究では、交流電場下に細胞を配置することで細胞に泳動力を生じさせる誘電泳動現象に着目し、この現象を用いた細胞アセンブリ技術と試料液の導入の精細な制御システムを複合化することで、多段階での細胞の集積化を可能とする細胞アセンブリシステムを構築した。さらに、開発されたシステムを用いて真皮および表皮細胞の多階層集積化と細胞チップ化を実施した。この細胞チップを7日間培養したところ、多層を成す皮膚構造が再構築された。アルコールを用いた刺激試験に対しても、刺激応答性が確認できた。以上、真皮・表皮細胞から構成される多階層培養皮膚エレメントの創製を実施し、薬効試験用の細胞チップとしての有効性を確認した。 The objective of this study was to reconstruct multilayered structure of micro organization using dielectrophoresis (DEP). DEP is a phenomenon in which a force is generated on a cell or particle when it is subjected to a non-uniform electric field. In this study, alginate-collagen micro-beads were developed for dielectrophoresis experiments and cell adhesive scaffold. Dermal and epi-dermal skin cells were assembled by DEP force to cover the beads and to construct multilayered structure. After the cell assembly, the cell-accumulated beads were cultured in vitro to reconstruct maultilayered skin tissue. Finally, the cell viability and proliferation of the cultured constructs were maintained during 7 days culture.
