内耳培養細胞HEI-OC1細胞を用いて、耳鳴内耳細胞モデル作製を試みた。まず、小胞体ストレスの細胞内誘導物質であるツニカマイシンをHEI-OC1細胞に暴露した。その後、control細胞とツニカマイシン処理細胞の間に有意差のある細胞生存率の条件(濃度100μg/ml、処理時間0-48h)を耳鳴内耳細胞モデルとした。この条件で、western blot法によるBDNFの発現は、時間依存性に低下することを確認した。さらに、この条件下で、内耳マーカーであるAtoh1も、時間依存性に低下することを確認した。 We try to establish the tinnitus model with inner ear cultured cell line HEI-OC1. At the beginning tunicamycin as endoplasmic reticulum stress was applied to HEI-OC1. After that inner ear tinnitus model was designed as the condition (concentration of 100 microgram/ml, and processing time of 0-48 hours), in which the cell validity was significantly different between the control and tunicamycin (+) condition. In this condition, the time dependant gradual reduction of appearance of BDNF was confirmed by western blot. In addition with this unique condition Atoh1, inner ear marker was also time dependently reduced.
