線虫の神経－行動相関を明らかにするため、新規なマイクロ流体デバイスと蛍光イメージング法を併用して調べる研究を行った。線虫頭部に層流を作り、温度または匂い物質などの濃度刺激を厳密に制御するマイクロ流体デバイスをデザインし、その時間的・空間的制御について検証を行った。またこのデバイス中の線虫行動を定量解析するためのソフトウェアを開発した。このデバイスを用いて、線虫頭部に高低2種類の匂い濃度刺激を加えたところ、線虫頭部が高濃度領域に長く滞在することを半自動的に定量することができた。またこの時、介在神経細胞AIYの神経活動に依存したカルシウム濃度上昇は頭部の屈曲行動と強く相関することを明らかにした。 Caenorhabditis elegans is known as one of model animals, and it has been used for understanding the relationship between neural functions and behaviors. The worm senses odors and shows taxis toward the source of favorite odor by detecting its concentration gradient. However, little is known how exactly worms can discriminate the concentration of odor and change their head turning behavior in regard to the response of olfactory neurons. For demonstrating the role of single neurons in specific behaviors, we investigated the neural activity from a single neuron by Ca and membrane potential imaging, and also quantitative behaviors by developing new types of micro-fluidic devices. We succeeded to visualize the Ca oscillation synchronized with the head movement during odor stimulation in AIY interneuron.
