本研究において、アミノ酸をはじめとする極性物質と脂質類をそれぞれメタボローム測定することとした。後者は、本研究を通じて前処理、測定条件の確立を進め、安定的な測定条件の確立によって血漿を用いて脂肪酸類、アシルカルニチン、酸化脂肪酸、胆汁酸類等が測定できる系をほぼ確立した。データ解析については、「健康の要因」として運動、飲酒、食事摂取を、「健康状態」と肝機能障害、高血圧、メタボリック症候群を取り上げ、CE-MS法で測定した血漿中代謝物との関連性を、交絡因子や多重比較性の調整を行いつつ検討した。その結果、飲酒、運動、蛋白質摂取のそれぞれに関連した血漿中のアミノ酸および関連代謝物質が同定された。 In this project, we developed metabolomics methodology for epidemiological setting both for CE-MS (polar) and LC-MS (lipids). In the statistical analysis, the association between health-related life style factors such as physical activity, alcohol drinking, diet intake and risk factors of non-communicable diseases including hepatopathy, hypertension, metabolic syndrome. As a result, various polar metabolites determined by CE-MS were determined with relation to alcohol drinking, hypertension, and physical activity. Branched-chain amino acids, leucine, isoleucine and valine, were significantly associated with degree of physical activity or clustering of metabolic syndrome. Further follow-up study is needed to elucidate whether these metabolites can be used for preventive biomarkers.
