本報告では, システムバイオロジー的概念に基づくオミクス解析から生命現象へのアプローチを利用した研究について学部中に行ったテーマである「分子クラウディング効果が大腸菌の中心炭素代謝反応に与える影響」, 「卵巣がんにおける薬剤耐性機構の解明に向けたプロテオーム及びリン酸化プロテオーム解析」の2つの内, 前者のみを述べる。細胞内の分子混雑状態が生体分子のふるまいに影響を与える分子クラウディング効果の一つとして, 酵素における熱安定性の向上が明らかにされている。分子混雑状態では, いくつかの酵素において, 熱に対する恒常性が増加すると先行研究で報告されているが, これらは単独の酵素反応のみであり, 代謝反応のような連続反応においては不明である。そこで本研究ではin vitroにて擬似的に再現した分子混雑状態での代謝反応をメタボローム解析から調べることで, 分子クラウディングが与える代謝反応への影響を明らかにすることを目指した。結果, ペントースリン酸経路の律速酵素であるGlucose 6-phosphate dehydrogenase(G6PDH)が分子クラウディング効果によって, 高温状態下でも活性を保ち, ペントースリン酸経路上流の反応を維持させることが示唆された。
