がん細胞が代謝をシフトしてATPや生体高分子の前駆体を生成するが, その理由や機序は謎であった。我々は, シースレスCE-MS法という高感度なメタボローム(全代謝産物)一斉分析法を開発し, トランスクリプトーム,プロテオームなどのシステム生物学的手法とともに, (遺伝性平滑筋腫症-腎細胞がん症候群)の原因であるフマル酸ヒドラターゼ遺伝子(FH)の変異を模倣した細胞株やノックアウトマウスを解析した。その結果, FH遺伝子の変異で蓄積したフマル酸が転写因子を安定化したり, 代謝酵素に結合したり, 代謝物に結合して代謝を制御していることを見出した。 Although cancer cells produce ATP and the precursors of macromolecules by switching metabolism, underlying mechanisms have been poorly understood. Here, we have developed a highly sensitive metabolome analysis method based on sheathless CE-MS device. This approach and together with systems biological technologies such as transcriptomics and proteomics were applied to fumarate hydratase (FH) knock out mouse and FH deficient cell lines, which mimic Hereditary leiomyomatosis and renal cell carcinoma that develops benign cutaneous and uterine leiomyomas, renal cysts and kidney cancer. Consequently, we found that accumulated fumarate caused by FH deletion regulated metabolism by activation transcription factor, inactivation of metabolic enzymes and conjugation to metabolites.
