CARS顕微鏡のC-Hモードで肺胞II型上皮細胞を観察すると非常に強いCARSシグナルを発するラメラ体と思われる構造体を観察した。肺胞II型上皮細胞をASEM用のディッシュ上で培養, 4%フォルムアルデヒドで固定し, ASEMでABCA3免疫染色の観察を行った。ABCA3は肺胞II型上皮細胞内でCARS顕微鏡での脂質と同様に細胞内に存在していた。今回の検討では, 残念ながら肺胞II型上皮細胞のASEM用のディッシュへの固着率が低く, 限られた検討しか行えなかった。ようやく, 液状コラーゲンを用いたコーティングで固着が安定出来るようになったので, 今後, 電子顕微鏡所見も含め, さらなる検討をしていく。 We observed lamellar body which emits very strong CARS signal in alveolar type II epithelial cells by C-H mode of CARS microscope. Primary cultured alveolar type II epithelial cells on a special dish for ASEM were fixed with 4% formaldehyde, immunostained with ABCA3 antibodies and observed by ASEM. ABCA3 was observed as similar to lipid observer by CARS microscopy in alveolar type II epithelial cells. Since fixed rate of alveolar type Ⅱ epithelial cells to a dish for ASEM was low unfortunately, only limited study was performed. Finally, because sticking with a coating using a liquid collagen began to be stable, we will continue to further consideration, including the electron microscope observations.
