7週齢CD1マウスより涙腺上細胞を培養し, 細胞株として樹立した。この涙腺上皮細胞の培養技術を用いて, 初代培養と樹立した細胞株から嚢胞様構造を効率よく形成させる条件を, 培地, 酵素処理時間, 培養時間から検討し, 最適条件を決定した。その条件において形成された嚢胞を用い, コヒーレント反ストークスラマン散乱顕微鏡(CARS顕微鏡)を用いて, 嚢胞内外の水の移動の観察, 解析を行なった。形成した嚢胞構造にサイズや構造のばらつきを認め, 分泌機能のアッセイ系として有用かどうかの評価には追加研究が必要である。 We established lacrimal gland epithelial cell line spontaneously from 7-week old CD1 mice. Cyst-like cell aggregates were generated with modifying condition of cultivated time, culture medium, and enzyme treating time. using the defined conditions form established cell line and primary culture. Those cyst were used to observe and analyze the water movement by Coherent Anti-Stokes Raman Scattering microscope. Although water movement could be observed, we need to generate cysts with the same size morphology for applying assay as the function of lacrimal glands. We need further study to solve this problems.
